La chromatographie liquide couplée à un spectromètre de masse en mode tandem avec source d’ionisation electrospray et analyseur triple quadripolaire est une technique très sensible et spécifique. Elle permet notamment la quantification des bases modidifées de l'ADN à un taux de l'ordre de 1 lésion par million de bases normales. Pour se faire, l'ADN est extrait des cellules ou des organes puis hydrolysé en monomères.

Dimerization of pyrimidine bases following absorption of UV photons leads a wide variety of photoproducts. A first class damage are cyclobutane pyrimidine dimers (CPD), produced through a [2+2] cycloaddition between the C5-C6 double bonds of two adjacent pyrimidine bases.
